【流式學(xué)習(xí)】γδT細(xì)胞：雖然我很少，但我很重要

T 細(xì)胞表面抗原受體（T cell receptor, TCR ）主要分為αβ TCR 和γδ TCR 兩大類，αβTCR細(xì)胞大家都很熟悉了，今天小達(dá)君主要跟大家分享一下γδT細(xì)胞。γδT細(xì)胞是一群固有免疫樣的T淋巴細(xì)胞，多數(shù)表型為 CD3⁺CD4^－CD8^－，主要分布在外周血和粘膜組織中，僅占人外周血 T 細(xì)胞的5%~10%?？刹灰】?/span>γδT細(xì)胞，在受到刺激活化后一樣能發(fā)揮多種效應(yīng)功能，包括殺傷感染的細(xì)胞和腫瘤細(xì)胞、分泌細(xì)胞因子和趨化因子、抗原遞呈功能及免疫調(diào)節(jié)功能。

γδT細(xì)胞的亞群：

Human：通常，人的γδT細(xì)胞根據(jù)其TCR基因重排過(guò)程中TCR δ鏈的不同主要分為兩大亞群：Vδ1T細(xì)胞亞群和Vδ2T細(xì)胞亞群，其中，與Vδ1T細(xì)胞對(duì)應(yīng)的TCRγ鏈主要為VγI基因家族成員（Vγ2/3/4/5/8），主要分布于黏膜和皮下組織中，在不同的組織中數(shù)量不等；Vδ2 T 細(xì)胞是γδ T 細(xì)胞的主要細(xì)胞群，其TCRγ鏈主要為VγII基因家族成員(Vγ9)，因此又叫 Vγ9Vδ2T 細(xì)胞，僅存在于靈長(zhǎng)類動(dòng)物和人體內(nèi)，主要分布于外周血中，占外周血淋巴細(xì)胞的0.5% ～10%。

Mouse：小鼠的γδT細(xì)胞由于其在體內(nèi)分布及功能特性的不同可分為不同的亞群。根據(jù)γδT細(xì)胞在發(fā)育過(guò)程中TCR基因重排時(shí)所選擇的Vγ鏈的不同，從結(jié)構(gòu)上可分為Vγ1-Vγ7，七個(gè)亞群。

活化與極化：

以人的γδT細(xì)胞為例，用不同的細(xì)胞因子以及磷酸化的抗原刺激后，靜息的γδT細(xì)胞可以極化為FoxP3⁺γδTreg、γδT17、γδTfh和γδT1，或成為活化的Vδ1T、Vδ2T細(xì)胞亞群。

功能：

γδT細(xì)胞具有多種功能，抗感染、抗腫瘤，在CAR-T治療中也發(fā)揮重要作用。

流式檢測(cè)：

以健康人的冷凍保存的PBMC為樣本，通過(guò)多色流式染色分析γδT細(xì)胞的分化階段。

抗體：

設(shè)門策略：

（A)

先通過(guò)FSC-A/SSC-A圈中淋巴細(xì)胞；
通過(guò)FSC-A/W排除粘連體；
通過(guò)SSC-A/W排除粘連體；
通過(guò)EMA死活染料排除掉死細(xì)胞；
圈中CD3^＋T淋巴細(xì)胞；
在CD3^＋T淋巴細(xì)胞中接著分析CD4^＋T細(xì)胞和CD8^＋T細(xì)胞的比例；因?yàn)槿送庵苎?/span>γδT細(xì)胞的表型基本都是CD4^－CD8^－，所以圈出CD4^－CD8^－雙陰性T細(xì)胞，準(zhǔn)備分析γδT表型；
圈中泛γδT細(xì)胞；
分析泛γδT細(xì)胞中的Vδ1^＋T細(xì)胞和Vδ2^＋T細(xì)胞 2個(gè)亞群的比例，以及Vδ1^－Vδ2^－T雙陰性細(xì)胞的比例；

(B)分析Vδ2^＋T細(xì)胞中CD27、CD28、CD45RA、CD16等標(biāo)記表達(dá)；

(C)分析Vδ1^＋T細(xì)胞中CD27、CD28、CD45RA、CD16等標(biāo)記表達(dá)。
